Our aim was to investigate the genetic diversity of 32 tamarind plus trees using inter-simple sequence repeat (ISSR) markers. Eighty four loci were examined using 12 markers, with a mean number of 4.42 loci per primer; 8 loci (9.52 %) were monomorphic and 76 (90.48 %) polymorphic, revealing genetic variability among the individuals. Polymorphic information content (PIC) values varied from 0.29 (ISSR_808) to 0.93 (ISSR_HB12), whereas the marker index ranged from to 26.4 (ISSR_814) to 62.5 (ISSR_17899A). Primers ISSR_HB11, ISSR_836, ISSR_842, ISSR_848, ISSR_860, ISSR_17899A and ISSR_17899B were useful to discriminate the grouping of the accessions according to their PIC values. Ward cluster analysis grouped accessions into two major groups with five subgroups with 46 % similarity according to Jaccard distance. The genotypes from Loja, Manabí and Guayas provinces were grouped in the first cluster; while only individuals from Manabí located in the other group, indicating major diversity in the latter province. Genotypes T1-ECUM-001 and T1-ECUM-002 presented 76 % similarity, while T1-ECUM-008, T1-ECUM-010, T1-ECUM-012, T1-ECUM-017 and T1-ECUM-018 shared 60 %. All materials from Loja grouped with 65 % similarity. Other genotypes clustered with similarity of 54 %. The cophenetic correlation coefficient (0.634) showed a good fit between the data matrix and the dendrogram results. A reasonable degree of diversity was found among tamarind genotypes potentially useful to select plus trees for clonal propagation as well as to identify diverse parents for hybridization programs.<hr/>Para aumentar el potencial del tamarindo como especie domesticada con valor económico es importante caracterizar la variabilidad en Ecuador con propósitos de mejoramiento. Nuestro objetivo fue investigar la diversidad genética de 32 árboles élite de tamarindo utilizando marcadores de secuencia inter-simple repetida (ISSR). Se examinaron 84 loci con 12 marcadores, con un número promedio de 4,42 loci per primer; 8 loci (9,52 %) fueron monomórficos y 76 (90,48 %) polimórficos, revelando variabilidad genética entre individuos. El contenido de información polimórfica (PIC) osciló entre 0,29 (ISSR_808) y 0,93 (ISSR_HB12), mientras que el índice de marcador fluctuó entre 26,4 (ISSR_814) y 62,5 (ISSR_17899A). ISSR_HB11, ISSR_836, ISSR_842, ISSR_848, ISSR_860, ISSR_17899A e ISSR_17899B fueron útiles para discriminar accesiones según sus CIPs. El análisis de conglomerados de Ward formó dos grupos principales y cinco subgrupos con 46 % de similitud según la distancia de Jaccard. Genotipos de Loja, Manabí y Guayas se aglomeraron en un grupo; mientras que sólo accesiones de Manabí quedaron en el otro, indicando mayor diversidad en la última provincia. Los genotipos T1-ECUM-001 y T1-ECUM-002 presentaron 76 % similitud, mientras T1-ECUM-008, T1-ECUM-010, T1-ECUM-012, T1-ECUM-017 y T1-ECUM-018 compartieron 60 %. Todos los materiales de Loja se agruparon con 65 % de similitud. Otros genotipos se concentraron con similitud de 54 %. El coeficiente de correlación cofenética (0,634) mostró buen ajuste entre la matriz de datos y los resultados del dendrograma. Se encontró un grado razonable de diversidad entre los genotipos de tamarindo potencialmente útil para seleccionar árboles élite para propagación clonal así como para identificar progenitores diversos para programas de hibridación. <![CDATA[<b>Attributes of quality in strawberry fruit 'Capitola' harvested in different climate condition in Venezuela</b>]]> http://hml-ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612017000300002&lng=pt&nrm=iso&tlng=pt La calidad de la fresa es afectada por las condiciones ambientales donde se desarrolla. Se estudiaron atributos fisicoquímicos de calidad del cultivar Capitola en tres localidades con diferentes condiciones climáticas determinadas por la altitud. Los frutos fueron cosechados en madurez organoléptica, uniformes en tamaño y coloración en Humocaro Bajo (Lara), Páramo de Cabimbú (Trujillo) y Colonia Tovar (Aragua). La calidad de los frutos mostró variabilidad entre las localidades, pero en general, presentaron una calidad fisicoquímica que se corresponde con los estándares de comercialización de la fresa. Los frutos de menor tamaño, pero de mayor firmeza fueron los de Humocaro Bajo y Colonia Tovar ubicadas a una menor altitud y con temperaturas superiores a las de Páramo de Cabimbú. En Humocaro Bajo con las mayores temperaturas, los frutos mostraron una coloración rojo brillante; en contraste con los de Páramo de Cabimbú que los frutos presentaron una coloración rojo oscuro. El mayor tamaño, así como la menor firmeza y acidez, junto a altos contenidos de sólidos solubles totales, antocianinas y vitamina C fueron encontrados en los frutos de Páramo de Cabimbú, lo cual sugiere que el comportamiento de estas variables estuvo condicionado por las bajas temperaturas y/o alta humedad relativa. En contraste, los mayores índices de color y contenidos de polifenoles totales se encontraron en los frutos de Humocaro Bajo. El análisis de componentes principales evidenció el efecto de las condiciones climáticas (altitud, temperatura, humedad relativa) sobre las variables fisicoquímicas de calidad de la fresa 'Capitola', al discriminarlas por localidad.<hr/>The quality of strawberry is affected by environmental conditions where the fruit develops. We evaluated physicochemical attributes at harvest of cultivar Capitola in three localities with different climatic conditions determined by the altitude. Fruits were harvested in organoleptic maturity, uniform in size and color in Humocaro Bajo (Lara), Páramo de Cabimbú (Trujillo) and Colonia Tovar (Aragua). The quality of the fruits showed variability among locations, although, in general, they presented a physicochemical quality that corresponds to the marketing standards. The fruits of smaller size, but greater firmness were those from Humocaro Bajo and Colonia Tovar located at lower altitude and higher temperatures, where the fruits showed a bright red color, in contrast to those from Páramo de Cabimbú, where fruits presented dark red color, larger fruits with less firmness. The largest size and lower firmness and acidity, as well as high content of total soluble solids, anthocyanins and vitamin, C were found in fruits from Páramo de Cabimbú, suggesting that the behavior of those variables were conditioned by low temperatures and high humidity. In contrast, the highest color indexes and total polyphenol contents were found in Humocaro Bajo fruits, where the highest temperatures were recorded. The principal component analysis showed the effect of weather conditions (altitude, temperature, relative humidity) on the quality variables of strawberry ‘Capitola’, which were discriminated by location. <![CDATA[<b>Estimation of combining ability and heterosis in non-conventional maize hibrids with high quality protein</b>]]> http://hml-ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612017000300003&lng=pt&nrm=iso&tlng=pt Con el objetivo de estimar los efectos de la actitud combinatoria general (ACG) y específica (ACE) y el nivel de heterosis, seis híbridos no convencionales de maíz con alta calidad de proteína (QPM) y sus progenitores fueron evaluados para rendimiento y otras variables de interés agronómico en Maracay, Venezuela. Los parámetros genéticos fueron estimados mediante el diseño dialélico de Griffing, método II, modelo I, bajo un bloque completo al azar, con tres repeticiones. Los resultados indicaron diferencias significativas (P≤0,05) entre genotipos para rendimiento, floración masculina y femenina, mala cobertura de la mazorca y pudrición de mazorca. El rendimiento medio de los híbridos no convencionales QPM fue de 6709,5 kg·ha-1, lo cual es 15 % superior a la media de sus progenitores (5810,0 kg·ha-1). El progenitor con mayor valor favorable de ACG para rendimiento, mala cobertura y mazorcas podridas fue S07TLYAB-2, mientras que los híbridos de mayor ACE para rendimiento fueron S07TLYAB-1/S07TLYAB-2 (7630,9 kg·ha-1), S07TLYAB-2/S03TLYQAB-05 (7605,3 kg·ha-1) y S07TLYAB-2/S03TLYQAB-03 (6693,6 kg·ha-1). Los dos primeros fueron los que presentaron mayor heterosis (≥23 %) y heterobeltiosis (≥15 %). Los resultados identifican variedades de maíz QPM que pueden ser incorporadas a los programas de mejoramiento genético para el desarrollo de híbridos no convencionales, poblaciones o generar líneas endocriadas con alta calidad de proteínas.<hr/>With the objective of estimating the general combining ability (GCA) and specific combining ability (SCA) effects and heterosis level, six non-conventional maize hybrids with high quality of protein (QPM), their inbred parents and two local checks were evaluated for grain yield and other agronomic traits at Maracay, Venezuela. The Griffing design, method I model II, was used to estimate GCA and SCA effects, on a complete randomized block design with three replications. The diallel analysis showed significant differences (P≤0.05) among genotypes for yield, days to anthesis, days to silking, uncovered maize ears and maize ear rot. Yield for non-conventional maize QPM hybrids was 6,709.5 kg·ha-1, 15 % higher than their inbred parents (5,810 kg·ha-1). S07TLYAB-2 showed the highest GCA for yield, uncovered maize ears and maize ear rot, and the crosses S07TLYAB-1/S07TLYAB-2 (7,630.9 kg·ha-1), S07TLYAB-2/S03TLYQAB-05 (7,605.3 kg·ha-1) and S07TLYAB-2/S03TLYQAB-03 (6,693.6 kg·ha-1) showed the highest SCA effects. The first two showed the highest mid-parent heterosis (≥23 %) and best parent heterosis (≥15 %) for yield. These results indicate that it is possible to develop a hybridization program using the best inbred parents to generate non-conventional hybrids, populations and segregating inbred lines with high quality protein. <![CDATA[<b>Effect of microfiltration and diafiltration on carotenoid and aromatic compounds of watermelon juice (<i>Citrullus lanatus</i> L.)</b>]]> http://hml-ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612017000300004&lng=pt&nrm=iso&tlng=pt Los carotenoides son un grupo de pigmentos naturales presentes en frutas y hortalizas con alta capacidad antioxidante, y además son precursores de la formación de aromas específicos. El objetivo del trabajo fue evaluar la distribución de carotenoides y compuestos aromáticos del jugo de sandía durante operaciones secuenciales de microfiltración y diafiltración. Los ensayos de microfiltración se realizaron en jugo con tratamiento enzimático previo, empleando una membrana de 0,8 μm a una presión promedio de 0,1 MPa, temperatura de 50 ºC y velocidad de flujo de 6,0 m·s-1, con 2 L de volumen de alimentación constante. Seguidamente se realizó un proceso de diafiltración del retenido utilizando agua potable. Se determinó la concentración de β-caroteno, isómeros trans y cis licopeno, sólidos solubles totales (SST), pH, acidez titulable, materia seca total y componentes aromáticos en las corrientes derivadas del proceso. El flujo promedio obtenido en la etapa de microfiltración fue de 114 L·h-1·m-2 a un factor de reducción volumétrica (FRV) de 10. La concentración de licopeno en el retenido luego de la microfiltración fue de 657±40 mg·kg-1. La etapa de diafiltración redujo los SST hasta 11,1±0,3 g·kg-1. El tratamiento enzimático no influyó en la composición de los compuestos aromáticos del jugo, sin embargo del proceso de microfiltración y diafiltración se obtuvieron corrientes con una distribución de compuestos aromáticos diferentes al jugo fresco.<hr/>Carotenoids are a group of natural pigments of fruits and vegetables with high antioxidant capacity, and are also precursors to the formation of specific aromas. The objective of this work was to evaluate the distribution of carotenoids and aromatic compounds of watermelon juice during sequential microfiltration and diafiltration operations. The microfiltration tests were performed in juice with previous enzymatic treatment, using a membrane of 0.8 μm at an average pressure of 0.1 MPa, temperature of 50 °C and flow velocity of 6.0 m·s-1, with 2 L constant feed volume. A retention diafiltration process was then performed using potable water. The concentration of β-carotene, trans and cis-lycopene isomers, total soluble solids (SST), pH, titratable acidity, total dry matter and aromatic components were determined in all streams derived from the process. The average flow obtained in the microfiltration stage was 114 L·h-1·m-2 at a volumetric reduction factor of 10. The concentration of lycopene in the retentate after the microfiltration was 657±40 mg·kg-1. The diafiltration step reduced the SST to 11.1±0.3 g·kg-1. The enzymatic treatment did not influence the composition of the aromatic compounds of the juice; however, from the microfiltration and diafiltration processes currents were obtained with a distribution of aromatic compounds different from the fresh juice. <![CDATA[<b>Characterization of a wild strain of <i>Saccharomyces cerevisiae </i>for obtaining a biopreparation with probiotic profile</b>]]> http://hml-ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612017000300005&lng=pt&nrm=iso&tlng=pt Generation of organic wastes is a worldwide environmental issue, although, if properly handled they may be a valuable source of animal nutrients. The objective was to determine the performance of a wild strain Saccharomyces cerevisiae in a biopreparation from ruminal content enriched with peels of fruits. Probiotic properties of the wild strain such as tolerance to bile salts, pH changes, changes in temperature, high concentrations of sodium chloride; additional to antagonism testing and gas production from glucose were verified in vitro. Microbial growth was evaluated in a medium prepared from clarified ruminal content 40 % v/v, enriched with peel wastes from papaya, pineapple and banana at 25, 50, and 75 % w/w concentrations. Results showed that wild strain had a higher growth in the medium obtained with 50 % w/v fruit wastes. Growth of the strain in this medium disclosed the highest biomass production at 40 hours (2.28x10(8) cfu·mL-1). The viability of the strain showed no important decrease during two months in the log scale of the inoculum. It is concluded that ruminal content enriched with fruit peels provides necessary nutrients for growth of the wild strain S. cerevisiae with probiotic characteristics and the biopreparation represents a supplement which may be useful not only at nutritional level but also to decrease environmental impact caused by these organic wastes.<hr/>La generación de desechos orgánicos es un problema medioambiental mundial, aunque, si se maneja adecuadamente, puede ser una valiosa fuente de nutrientes para animales. El objetivo fue determinar el rendimiento de una cepa silvestre de Saccharomyces cerevisiae en un biopreparado a partir de contenido ruminal enriquecido con cáscaras de frutas. Se verificaron in vitro las propiedades probióticas de la cepa como tolerancia a sales biliares, cambios de pH, cambios de temperatura y altas concentraciones de cloruro de sodio, así como de pruebas de antagonismo y producción de gas a partir de la glucosa. Se evaluó el crecimiento microbiano en el medio elaborado a partir del contenido ruminal al 40% v/v, enriquecido con desechos de cáscaras de papaya, piña y banano en concentraciones de 25, 50 y 75 %. El mayor crecimiento se produjo en el medio con 50 % de desechos de frutas y la cepa reveló la mayor producción de biomasa a las 40 horas (2,28x10(8) ufc·mL-1). Su viabilidad no presentó disminución importante en la escala logarítmica del inóculo durante dos meses. Se concluye que el contenido ruminal enriquecido con cáscaras de frutas al 50 %, proporciona los nutrientes necesarios para el crecimiento de S. cerevisiae con características probióticas, y el biopreparado representa un suplemento que no solo puede ser benéfico a nivel nutricional, sino que también disminuiría el impacto ambiental ocasionado por estos desechos orgánicos. <![CDATA[<b>Potentialities and limitations of agricultural soils in Lara State, Venezuela</b>]]> http://hml-ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612017000300006&lng=pt&nrm=iso&tlng=pt El Departamento de Química y Suelos de la Universidad Centroccidental Lisandro Alvarado, en el centro occidente de Venezuela, posee una de las bases de datos más importante de los suelos de la región. Sin embargo, la misma no ha sido empleada para identificar los factores edáficos que afectan la producción agrícola. El objetivo de este trabajo fue identificar las principales limitaciones y potencialidades de suelos de una de las principales zonas agrícolas del estado Lara. Para ello se analizaron datos de pH, conductividad eléctrica (CE) materia orgánica (MO), contenidos de arcilla, limo, arena, fósforo, calcio, magnesio, potasio y aluminio, correspondientes a un periodo de 20 años. Los datos fue georreferenciados, tomando como base un mapa 1:100.000 y ubicando en el mismo la zona baja de la depresión de Quíbor y la zona de piedemonte. A partir de estudios previos se construyeron categorías de calidad de suelos para la clasificación de cada una de las variables evaluadas y se realizó un análisis de componentes principales para estudiar la distribución de las muestras en función de dichas variables. Los resultados señalan que los suelos de las zonas de piedemonte presentaron valores altos de fósforo y materia orgánica, siendo sus principales limitaciones, bajos valores del pH y altos valores de aluminio cambiable, mientras que los suelos de las zonas bajas, presentaron una alta disponibilidad de nutrientes, pero limitaciones por los altos valores de limo, arcilla y CE, los cuales conllevan a problemas de encostramiento, sellado superficial, erosión y afectación por sales.<hr/>The Chemistry and Soil Department of the Universidad Centroccidental Lisandro Alvarado, in the center western of Venezuela, has one of the most important soil database of the region; however, it has not been used to identify the edaphic factors affecting agricultural production. The objective of this study was to identify main soil constraints and potentials from the principal agricultural areas of the Lara State. Data from pH, electrical conductivity (EC), organic matter (OM), content of clay, silt, sand, phosphorus, calcium, magnesium, potassium and aluminum of 20 years period were analyzed. The data were georeferenced on a 1:100,000 base map and locating on it, the Quibor depression plain and the piedmont areas. By using previous studies, soils quality categories were developed for classification of each of the evaluated variables and an analysis of their main components was performed, to study the samples distribution as a function of those variables. Results indicate that soils of piedmont areas presented high values of P and OM, being its main limitation low pH and high Al values, while soils from low areas, presented a high nutrient availability, but limitations resulting from their high silt and clay content, and EC values, which become on problems of surface sealing, crusting, erosion and salt affectation. <![CDATA[<b>Genetic variability of peaches cultivated in ‘El Peñón de Gabante’, Aragua State, Venezuela</b>]]> http://hml-ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612017000300007&lng=pt&nrm=iso&tlng=pt En este estudio se planteó la identificación molecular de durazneros cultivados en Venezuela, información que podría ser empleada como base para programas de mejoramiento genético. Hasta hoy sólo ha sido usada la clasificación visual, la cual puede ser subjetiva. El presente trabajo se realizó para caracterizar la variabilidad genética en Prunus persica (L.) Batsch cultivado en una de las principales zonas de producción venezolana. Se colectaron hojas jóvenes completamente desarrolladas de diferentes cultivares en huertos comerciales del Peñón de Gabante, estado Aragua. Se utilizaron 10 pares de iniciadores moleculares microsatélites (SSR). La evaluación del perfil electroforético se realizó por medio del índice de similaridad. Se realizaron análisis de clasificación y ordenación de las muestras. El dendrograma se construyó usando el método de grupos-pares no ponderados con promedios aritméticos (UPGMA). Se confirmó la estrecha base genética que existe dentro de la especie muestreada. Fue posible discriminar molecularmente los cultivares y esa discriminación correspondió a la realizada por los productores.<hr/>In this study the molecular identification of peaches cultivated in Venezuela was proposed, information that could be used as a basis for breeding programs. To date, only visual classification has been used, which can be subjective. The present work was carried out to characterize genetic variability in Prunus persica (L.) Batsch cultivated in one of the main Venezuelan production zones. Fully-developed young leaves of different cultivars were collected in commercial orchards. Ten pairs of primers (SSR) were used. Evaluations of the electrophoretic profiles were performed using the similarity index, along with classification and ordering analyzes of the samples. Dendrogram was constructed using the unweighted paired-arithmetic average (UPGMA) method. A close genetic basis within the peaches cultivars was confirmed, and it was possible to discriminate molecularly cultivars, which fairly corresponded to those identified by the producers. <![CDATA[<b>Effect of the freeze-drying on the physico-chemical characteristics of rocoto chili pepper (C<i>apsicum pubescens </i>R & P)</b>]]> http://hml-ve.scielo.org/scielo.php?script=sci_arttext&pid=S1316-33612017000300008&lng=pt&nrm=iso&tlng=pt Con la finalidad de fomentar la transformación agroindustrial en Capsicum pubescens, en esta investigación se deshidrató ají rocoto con y sin semillas, cultivado en la zona rural de San Cristóbal, Medellín, Antioquia (Colombia), mediante liofilización y se evaluó su efecto sobre las características del producto final. Se empleó un arreglo de parcelas dividas, con el factor de la programación de la velocidad de calentamiento, aplicando una tasa de aumento de temperatura entre 0,02°C/min a 0,05°C/min durante la sublimación en la parcela principal, y el factor semilla en la subparcela, con una aleatorización completamente al azar y tres repeticiones. Se evaluaron las características físico-químicas del producto final de contenido de humedad, actividad de agua (a w), pH, acidez, indicadores de color en el sistema CIE-L*a*b*y capacidad de rehidratación, así como posible afectación del tejido usando microscopía electrónica de barrido. El porcentaje de humedad, a w, pH y acidez no se afectaron significativamente entre los tratamientos liofilizados (P>0,05) logrando obtener valores máximos de 8,62% de humedad y a w de 0,33. Se mejoró la luminosidad (L*) y cromaticidad (C*) del ají rocoto liofilizado con y sin semilla con respecto al producto en fresco; además, se aumentaron los atributos de color b* y h°. Con la velocidad de menor calentamiento para la liofilización, los tratamientos con y sin semillas presentaron una mayor capacidad de rehidratación. En los frutos sin semilla se necesitó menor tiempo para alcanzar tanto las temperaturas de congelación como de sublimación, a la vez que mejoraron la mayoría de los indicadores de color.<hr/>In order to foment the agroindustrial transformation of Capsicum pubescens, in this research we dehydrated rocoto chili peppers with and without seeds, harvested in the rural zone of San Cristobal Medellin, Antioquia (Colombia), through freeze-drying, and the properties in the final product were evaluated. Three times of the heating rate were programmed (temperature rise from 0.02 to 0.05 °C per minute during sublimation) using fruits with and without seeds. A split plot design was used with the programming in the main plot, and the presence of seeds in the subplot in a randomized block design with three replications. It was evaluated the physico-chemical properties of milled final product characteristics, moisture content (%), water activity (a w), pH, acidity, color indicators CIE-L, and rehydration capacity (RC), as well as possible changes of the plant tissue using scanning electron microscopy. The moisture percentage, a w, pH and acidity, did not differ among treatments (P≤0.05), reaching maximum moisture of 8.62 % and a w of 0.33. The luminosity (L*) and chroma (C*) of the dried with respect to fresh fruit was improved. Attributes of color b* and h° increased as well. With the lower heating rate for freeze drying, the fruit with and without seeds presented higher RC. The seedless fruits required less time to reach both freezing and sublimation temperatures and showed higher values of most color indicators.
